Thu Duc City, March 17", 2023 Student’s signature Luu Khai Nhien 1 ABSTRACT The aim of this study was to investigate the effect of fetal bovine serum (FBS) supplemented in culture medium for fibroblast cells derived from porcine ovarian tissue. Tissue explants were submitted to primary culture in medium with different FBS concentrations for 8 days with a medium change at day 4. Results showed that between the two treatments of supplementing 10 and 20% FBS throughout 8 days, a significant difference (5.05) of cells harvested was found. No differences in harvested cell numbers were observed between supplementing 10 — 20% and 20 — 10% at first 4 days - later 4 days (5.
Three levels of FBS concentration (10, 15, and 20%) were evaluated for the subculture medium through a growth curve and population doubling trme (PDT). There were significant differences between supplementations of 10 and 20% FBS on day 3 (4.45, respectively), and day 7 (4. No differences were recorded between the 15 and 20% treatments. The PDT of 10, 15, and 20% treatments were 68.
The growth curve with the exponential phase spanning from day 1 to day 5 was determined. In conclusion, supplementation of 10% — 20% FBS in first 4 days — later 4 days of primary culture medium and subculture in medium with 15% FBS enhanced the total number of fibroblast cells isolated and proliferated derived from porcine ovarian tissue. Problem statement Isolation and proliferation of fibroblast cells are the first and crucial steps in generating materials for many experiments on cell physiology, cryopreservation, and somatic cell nuclear transfer (SCNT).
In many established protocols for fibroblast cells culture, fetal bovine serum (FBS) is considered an indispensable ingredient in the culture medium (Reuisi ef a/, 2009; Hu et ai, 2012; Phelan and May, 2015; Mestre- Citrinovitz et al, 2016; Bryja et al, 2020). Since its discovery, FBS had been proven to possess many important factors for the in vitro growth and proliferation of cells derived from animals (Carrel and Ebeling, 1922; Treadwell and Ross, 1963). Fibroblast cells are mostly found in connective tissue with the ability to grow vigorously (Tran Thi Khanh Hoa, 2018). Fibroblast cells are also responsible for the formation of collagens, proteoglycans, fibronectin, and other proteins in the extracellular matrix (Lu ef a/, 2011).
When cultured in vitro, fibroblast cells express monolayer properties, which is utilized as feeder layers for co-culture procedures with different types of cell (Glass ef al, 1979; Kuzan and Wright, 1981; Kuzan and Wright, 1982; Allen and Wright, 1984; Yeoman ef a/, 2005). Chinese hamster ovary (CHO) cell line is used as a model for research, utilizing cells isolated from ovarian tissue (Omasa ef al, 2010). Hence, the evaluation of FBS when supplemented at various concentrations to the capability of isolation and proliferation of cells derived from porcine ovarian tissue is needed. Objectives To assess the optimal concentration of fetal bovine serum supplemented in primary or secondary culture medium to maximize the quantity of fibroblast cells collected after culturing and proliferating.

Animal cells Animal cells are found in mammals, including humans, and are a type of eukaryotic cell. These cells are distinct for their absence of a cell wall and their possession of organelles such as mitochondria and lysosomes.
In animals, there are four main types of tissue: epithelial, connective, muscular, and nervous. Epithelial tissue covers the body's surface and lines internal organs, consisting of tightly packed cells that create a barrier between the internal and external environment. Connective tissue offers support and connects different parts of the body, comprised of various cells including fibroblast cells and extracellular matrix elements such as collagen and proteoglycans. Muscular tissue is responsible for movement, divided into three categories: skeletal, smooth, and cardiac muscle.
The skeletal muscle controls voluntary movement, the smooth muscle controls involuntary movement, and the cardiac muscle powers blood circulation. Nervous tissue transmits and processes signals in the body, made up of neurons and glial cells. Neurons transmit signals and glial cells provide support and protection for neurons. Ovary and ovarian tissue The ovary is a part of the female reproduction system in mammals, responsible for the production of ova and secretion of endocrines.
In pigs, the ovaries are located in the lower abdomen area along with oviducts and uterine horns (Figure 2. On the surface, the ovary is covered by the germinal epithelium while the tunica albuginea and stroma (ovarian cortex and medulla) make up the internal structure. The ovarian cortex contains ovarian follicles, fibroblast cells, collagen, and elastic fibers. Nerves, blood, and lymphatic vessels reside in the ovarian medulla.
In mice, tissue from Chinese hamster ovary (CHO) is used in cell culture to produce a reliable model for research and production purposes (Omasa ef a/, 2010). Since fibroblast cells can originate from the ovarian cortex, this organ is suitable for the primary cultivation of fibroblast cells. In other notes, pig ovaries are considered waste in the meat industry, thus the recovery of the said organ is undeniably easy and cheap. Animal cell culturing Cell culture involves the growth of cells, either taken from animal tissue or whole animals, by providing them with essential nutrients and growth factors. Cell culture enables individual cells to behave as standalone units, similar to microorganisms like bacteria or fungi. These cells can divide via mitosis, leading to a growing population until limited by factors such as nutrient depletion.
Cell cultures usually consist of cells of a single type, which can be either genetically identical, forming a homogeneous population, or show some genetic variation, forming a heterogeneous population (Butler, 2004).
