I. Tổng quan về đồ án chế biến bã đậu nành lên men bằng SSF

Đồ án nghiên cứu chế biến bã đậu nành (SBM) thành sản phẩm lên men bằng phương pháp lên men rắn (SSF) nhằm cải thiện giá trị dinh dưỡng. Các ANFs chính bao gồm trypsin inhibitor (ức chế enzyme tiêu hóa protein), lectin (gây tổn thương niêm mạc ruột), phytate (giảm hấp thu khoáng chất) và oligosaccharides (gây đầy hơi). SBM cũng có tỷ lệ protein tiêu hóa thấp do cấu trúc protein bị biến tính trong quá trình ép dầu. Vi khuẩn Bacillus spp. (điển hình là Bacillus velezensis) được lựa chọn do khả năng sản xuất enzyme protease mạnh, phân hủy protein thành peptide và axit amin. Quá trình SSF diễn ra trong 3-7 ngày, với điều kiện tối ưu về nhiệt độ (30-37°C), pH (6.5-7.5) và tỷ lệ giống (5-10%). II. Những thách thức trong xử lý bã đậu nành bằng phương pháp truyền thống

Các phương pháp xử lý truyền thống như hấp, nướng hoặc ép dầu không đủ để phá hủy hoàn toàn trypsin inhibitor (chỉ giảm 50-70%) và phytate (còn lại 60-80%). Hơn nữa, quá trình nhiệt độ cao có thể làm biến tính protein, giảm giá trị sinh học. 2.1. Hạn chế của phương pháp nhiệt độ cao

Phương pháp nhiệt độ cao (hấp, nướng) chủ yếu nhằm phá hủy trypsin inhibitor nhưng không hiệu quả đối với phytate. Nhiệt độ cao (120-130°C) trong 15-30 phút chỉ giảm 30-50% phytate, đồng thời gây biến tính protein (Maillard reaction), giảm khả năng tiêu hóa axit amin thiết yếu như lysine. Ngoài ra, quá trình nhiệt có thể sinh ra các hợp chất độc hại (ví dụ: acrylamide) nếu nguyên liệu chứa carbohydrate khử. Do đó, phương pháp này không phù hợp cho sản xuất thức ăn chăn nuôi an toàn, đặc biệt là đối với vật nuôi non nhạy cảm.

2.2. Khó khăn trong tối ưu hóa SSF

III. Phương pháp nghiên cứu chế biến bã đậu nành lên men bằng SSF

Nghiên cứu sử dụng chủng Bacillus velezensis được phân lập từ môi trường tự nhiên, có khả năng sản xuất protease mạnh. SBM được xử lý tiền ủ (ngâm 30 phút ở 50°C) trước khi bổ sung 10% giống vi khuẩn (10^8 CFU/g). Quá trình SSF diễn ra trong 5 ngày ở 35°C, độ ẩm 55%. Protein được đo bằng phương pháp Lowry, carbohydrate bằng phương pháp Phenol-Sulfuric Acid. Đánh giá hiệu quả thông qua tỷ lệ giảm trypsin inhibitor (sử dụng phương pháp Kunitz), hàm lượng protein tiêu hóa (in vitro) và khả năng hấp thu khoáng chất (phytase activity). Thí nghiệm được lặp lại 3 lần với điều kiện đối chứng (SBM không lên men). Kết quả cho thấy hàm lượng protein tăng từ 48% (SBM thô) lên 53.58% (sau SSF), đồng thời trypsin inhibitor giảm 85%. Sản phẩm lên men đạt tiêu chuẩn thức ăn chăn nuôi (hàm lượng protein >50%, ANFs <5%).

3.1. Quy trình chuẩn bị nguyên liệu

SBM thô được lấy từ nhà máy sản xuất dầu đậu nành tại Đồng Nai, sàng lọc loại bỏ tạp chất (đá, kim loại) và nghiền thành bột mịn (kích thước 0.5-1 mm). Nguyên liệu được tiệt trùng ở 121°C trong 15 phút để tiêu diệt vi sinh vật có hại. Bổ sung 5% bột ngô làm chất nền phụ nhằm tăng cường độ xốp, tạo điều kiện thuận lợi cho vi sinh vật phát triển. Dung dịch đệm phosphate (pH 7.0) được sử dụng để điều chỉnh độ ẩm về 55%. Giống vi khuẩn Bacillus velezensis được hoạt hóa trong môi trường LB lỏng (37°C, 180 rpm) trong 12 giờ trước khi bổ sung vào SBM với tỷ lệ 10% (v/w).

3.2. Phương pháp đánh giá hiệu quả lên men

Hiệu quả SSF được đánh giá thông qua 3 chỉ tiêu chính: hàm lượng protein (Lowry), tỷ lệ giảm trypsin inhibitor (Kunitz) và hoạt độ phytase. Protein tiêu hóa được đo bằng phương pháp pepsin-pancreatin in vitro (37°C, 4 giờ). Phytase activity được xác định bằng phương pháp đo giải phóng phosphate vô cơ từ phytate (sử dụng dung dịch Na-phytate 0.005M). Kết quả được so sánh với SBM không lên men và SBM xử lý nhiệt. Ngoài ra, sản phẩm lên men được phân tích vi sinh (đếm khuẩn lạc trên đĩa thạch) để đảm bảo không nhiễm nấm mốc. Kết quả thí nghiệm được xử lý bằng phần mềm SPSS (ANOVA, p<0.05).

IV. Kết quả nghiên cứu và ứng dụng của bã đậu nành lên men

Sau 5 ngày SSF, sản phẩm bã đậu nành lên men (FSBM) đạt hàm lượng protein 53.58%, cao hơn 11.6% so với SBM thô. Trypsin inhibitor giảm 85% (từ 12.5 TIU/mg xuống 1.8 TIU/mg), phytate giảm 60% (từ 1.2% xuống 0.48%). Protein tiêu hóa in vitro tăng từ 78% (SBM thô) lên 92% (FSBM). Hoạt độ phytase đạt 0.8 U/g, cải thiện đáng kể khả năng hấp thu khoáng chất. FSBM có thể thay thế 30-50% protein động vật trong thức ăn chăn nuôi heo con giai đoạn cai sữa, giúp tăng trọng lượng 15% sau 4 tuần sử dụng. Sản phẩm cũng có thể ứng dụng trong sản xuất thức ăn thủy sản (cá tra, tôm) do hàm lượng protein dễ tiêu hóa cao. It contains many nutrients suitable for animal feed production. In addition to being a high-protein product, SBM (soybean meal) also contains significant amounts of anti-nutritional factors, and those elements need to be removed to increase the absorption of SBM. Introduction of Soybean Meal Soybean meal (SBM) is one of the essential protein sources used in the poultry and livestock industry, accounting for about two-thirds of the total protein feed production worldwide.

The protein value in SBM is higher than that of other plant protein sources and is considered a benchmark against which other protein sources are compared [1]. Soybean meal is a by-product of oil extraction. Its crude protein content often classifies SBM for marketing purposes. Soybeans, from the hulled seeds, the high-protein varieties have been harvested and contain 47-49% protein and 3% crude fiber, while another soybean meal contains less than 47% protein and more than 6% fiber, including the shell or part of the cover [2].

Soybean meal (or okara) is the fibrous part of beans, the insoluble part of soybeans left after soybeans have been pureed and filtered to produce soy milk and tofu. Generally, they are white or yellowish and are rich in nutrients, including insoluble and soluble fiber, protein, and other minerals (such as calcium, potassium, and niacin). Most of the isoflavones, B vitamins, and fat- soluble nutritional elements (including linoleic acid, soy lecithin, phytosterols, linolenic acid, vitamin D, and tocopherol) present in soybeans are still present in the SBM [3]. Several anti-nutritional factors are present in the SBM, such as saponins, trypsin inhibitors, and hemagglutinin.

SBM fermentation can aid in digestion and absorption of nutrients and improve nutritional value. It clears the smell of beans and increases the amount of edible fiber, fatty acids, sugars, free amino acids, vitamin B2, vitamin B12, and flavoproteins. SBM is a functional food to prevent diabetes, hyperlipidemia, and obesity. However, due to its high protein, soluble and insoluble fiber content, and high perishability, SBM is usually used immediately or cooked and frozen for storage.

The most attractive point of SBM that can be mentioned is the low price and low calories. More recently, okara has been used as an ingredient in cookies, cakes, and other confectionery, as well as in traditional side dishes. Their low-calorie composition makes them an ideal diet food [4]. SBM is the most popular soybean product in animal diets, with other products being used to varying degrees.

These products include full-fat soybeans, soy protein concentrate (SPC), soybean oil soy protein isolate (SPI), and soybean hulls. Each of these products has unique nutritional properties that make them suitable for inclusion in the diets of certain animals. In particular, in the composition of SBM, amino acids (AA) have supplemented the AA components of different grains with the practical use of AA in soybean meal, also explaining why in the food industry. For pet food, soybean meal is more and more widely used [5].
