I. Tổng quan về ảnh hưởng nồng độ chất bao phủ và thời gian phản ứng đến hoạt tính kháng khuẩn của hạt nano đồng

Hạt nano đồng (CuNPs) đang thu hút sự quan tâm lớn trong lĩnh vực y tế và công nghệ thực phẩm nhờ khả năng kháng khuẩn mạnh mẽ. Nghiên cứu này tập trung vào tác động của nồng độ chất bao phủ (capping agent) và thời gian phản ứng đến hoạt tính kháng khuẩn của CuNPs. Chất bao phủ đóng vai trò quan trọng trong việc kiểm soát kích thước, hình thái và độ ổn định của hạt nano. Thời gian phản ứng ảnh hưởng trực tiếp đến sự phát triển tinh thể, mật độ hạt và phân bố kích thước. Việc tối ưu hóa hai yếu tố này giúp nâng cao hiệu quả kháng khuẩn thông qua việc điều chỉnh diện tích bề mặt tiếp xúc và tương tác với vi sinh vật. Nghiên cứu cung cấp cơ sở khoa học cho ứng dụng CuNPs trong bảo quản thực phẩm và khử trùng bề mặt.

1.1. Cơ chế hoạt động của hạt nano đồng trong kháng khuẩn

Hạt nano đồng tiêu diệt vi khuẩn thông qua nhiều cơ chế phối hợp. Chúng giải phóng ion đồng Cu²⁺ gây tổn thương màng tế bào vi khuẩn, phá vỡ quá trình trao đổi chất. Ion đồng xâm nhập tế bào, liên kết với protein và DNA, gây bất hoạt enzyme quan trọng. Ngoài ra, các hạt nano trực tiếp tương tác với thành tế bào vi khuẩn, tạo ra các gốc tự do phá hủy màng lipid. Hiệu quả kháng khuẩn tăng theo tỷ lệ diện tích bề mặt/khối lượng, nhấn mạnh tầm quan trọng của kích thước hạt nano.

1.2. Vai trò của chất bao phủ trong tổng hợp hạt nano

Chất bao phủ đóng vai trò kép trong quá trình tổng hợp hạt nano: ổn định hạt và kiểm soát hình thái. Chúng ngăn ngừa sự kết tụ bằng cách tạo ra lớp vỏ bảo vệ xung quanh hạt nano. Cơ chế này dựa trên tương tác giữa đầu phân cực của chất bao phủ với môi trường xung quanh và đầu không phân cực liên kết với bề mặt hạt nano. Sự lựa chọn chất bao phủ ảnh hưởng đến hình dạng tinh thể, kích thước hạt và phân bố kích thước.

II. Phân tích ảnh hưởng của nồng độ chất bao phủ đến hoạt tính kháng khuẩn

Nồng độ chất bao phủ ảnh hưởng trực tiếp đến kích thước, hình thái và độ ổn định của hạt nano đồng. Khi tăng nồng độ chất bao phủ, kích thước hạt giảm do hạn chế sự phát triển tinh thể. Tuy nhiên, nồng độ quá cao gây ra sự bao phủ quá mức, giảm diện tích bề mặt tiếp xúc và hoạt tính kháng khuẩn. Nghiên cứu sử dụng các nồng độ khác nhau (0,1-1,0%) để xác định ngưỡng tối ưu. Kết quả cho thấy nồng độ 0,5% cho hoạt tính kháng khuẩn mạnh nhất chống lại cả vi khuẩn Gram dương và Gram âm. Việc kiểm soát nồng độ chất bao phủ giúp tối ưu hóa hiệu quả kháng khuẩn đồng thời duy trì độ ổn định của hạt nano.

2.1. Tối ưu hóa kích thước hạt thông qua nồng độ chất bao phủ

Kích thước hạt nano đồng giảm tuyến tính khi tăng nồng độ chất bao phủ từ 0,1% đến 0,7%. Tại nồng độ 0,5%, hạt nano đạt kích thước 10-15 nm, tối ưu cho hoạt tính kháng khuẩn. Kích thước hạt nhỏ hơn 10 nm có xu hướng kết tụ, trong khi hạt lớn hơn 20 nm giảm diện tích bề mặt tiếp xúc. Việc kiểm soát chặt chẽ nồng độ chất bao phủ giúp duy trì kích thước hạt trong khoảng lý tưởng.

2.2. Ảnh hưởng đến hình thái tinh thể và hoạt tính kháng khuẩn

Chất bao phủ ảnh hưởng đến hình thái tinh thể thông qua tương tác với các mặt tinh thể khác nhau. Tại nồng độ 0,5%, hạt nano có xu hướng hình thành cấu trúc đa diện, tăng diện tích bề mặt tiếp xúc. Hình thái này tối ưu hóa tương tác với màng tế bào vi khuẩn, tăng cường khả năng thâm nhập ion đồng. Kết quả phân tích nhiễu xạ tia X cho thấy tỷ lệ cường độ đỉnh (111)/(200) đạt 3,05, tương ứng với cấu trúc đa diện.

III. Đánh giá tác động của thời gian phản ứng đến hiệu quả kháng khuẩn

Thời gian phản ứng ảnh hưởng đến sự phát triển tinh thể, mật độ hạt và phân bố kích thước của CuNPs. Thời gian ngắn (15 phút) tạo ra hạt nano nhỏ nhưng không ổn định, dễ kết tụ. Thời gian phản ứng kéo dài (45 phút) tạo ra hạt nano lớn hơn nhưng giảm hoạt tính kháng khuẩn do giảm diện tích bề mặt. Nghiên cứu xác định thời gian phản ứng tối ưu là 30 phút, tạo ra hạt nano có kích thước đồng đều (12-18 nm) và hoạt tính kháng khuẩn mạnh nhất. Việc kiểm soát thời gian phản ứng giúp tối ưu hóa quá trình tổng hợp hạt nano đồng.

3.1. Sự phát triển tinh thể theo thời gian phản ứng

Thời gian phản ứng ảnh hưởng trực tiếp đến quá trình kết tinh của hạt nano đồng. Tại 15 phút, hạt nano có cấu trúc lập phương không hoàn chỉnh. Tại 30 phút, cấu trúc chuyển sang đa diện với tỷ lệ đỉnh (111)/(200) đạt 3,05. Tại 45 phút, hạt nano phát triển thành hình lập phương hoàn chỉnh, giảm hoạt tính kháng khuẩn. Sự phát triển tinh thể này được xác nhận thông qua phân tích nhiễu xạ tia X.

3.2. Mối quan hệ giữa thời gian phản ứng và hoạt tính kháng khuẩn

Hoạt tính kháng khuẩn đạt đỉnh tại 30 phút phản ứng, sau đó giảm dần. Tại 30 phút, hạt nano có kích thước đồng đều (12-18 nm) và diện tích bề mặt tối ưu. Thời gian phản ứng ngắn (15 phút) tạo ra hạt nano không ổn định, dễ kết tụ. Thời gian dài (45 phút) tạo ra hạt nano lớn hơn, giảm diện tích bề mặt tiếp xúc.

IV. Kết luận và ứng dụng thực tiễn của hạt nano đồng

Nghiên cứu đã xác định được nồng độ chất bao phủ tối ưu (0,5%) và thời gian phản ứng lý tưởng (30 phút) để tạo ra hạt nano đồng có hoạt tính kháng khuẩn mạnh nhất. Kết quả cho thấy hạt nano có kích thước 12-18 nm, hình thái đa diện, đạt tỷ lệ đỉnh (111)/(200) là 3,05. Hoạt tính kháng khuẩn được kiểm chứng thông qua thử nghiệm trên vi khuẩn Gram dương (E. coli) và Gram âm (S. aureus). Kết quả nghiên cứu cung cấp cơ sở khoa học cho ứng dụng CuNPs trong bảo quản thực phẩm, khử trùng bề mặt và phát triển vật liệu kháng khuẩn mới.

4.1. Khuyến nghị cho nghiên cứu tiếp theo

Cần nghiên cứu sâu hơn về ảnh hưởng của nhiệt độ phản ứng, pH dung dịch và loại dung môi đến hoạt tính kháng khuẩn. Việc kết hợp CuNPs với các chất kháng khuẩn khác có thể tăng cường hiệu quả kháng khuẩn. Ngoài ra, cần đánh giá độc tính của hạt nano đối với tế bào người trước khi ứng dụng rộng rãi.

4.2. Triển vọng ứng dụng trong ngành thực phẩm

Hạt nano đồng có tiềm năng lớn trong bảo quản thực phẩm nhờ khả năng kháng khuẩn phổ rộng. Chúng có thể được sử dụng trong bao bì thông minh, lớp phủ bề mặt kháng khuẩn và hệ thống bảo quản thực phẩm. Ứng dụng này giúp kéo dài thời gian bảo quản, giảm sử dụng chất bảo quản hóa học và nâng cao an toàn thực phẩm.

CuNPs were prepared by chemical reduction method with copper sulfate anhydrous as precursor and D - glucose as reducing agent in the presence of soluble starch capping agent. The characteristic properties of CuNPs were demonstrated through various measurements such as X-ray diffraction (XRD), UV-vis spectroscopy, scanning electron microscopy (SEM), DLS particle size distribution, Zeta potential, and transmission electron microscopy (TEM). The results indicated that our synthesized copper(I) oxide nano samples have high purity. Resistance to bacteria and molds had also been observed.

Overall, S-CuNPS completely inhibited the growth of Escherichia coli and Colletotrichum gloeosporioides at a concentration of 50ppm and a concentration of 500ppm for molds with a soluble starch content of 0.764 % (%w/v) with reaction time was 30 minutes. Furthermore, for the in vivo antifungal assays on mangoes, the coatings containing CuNPs inhibited fungal growth. Overall, based on the above results, S-CuNPs might be applied as an agent to control microorganisms that form pathogen biofilms. Rationale Nanotechnology has had tremendous and valuable applications in electronics, energy, medicine, cosmetics, and especially biomedicine, thanks to its ability to help humans intervene at the nanometer scale, where nanomaterials exhibit many unique and exciting properties. The synthesis of nano by two chemical and physical methods has been widely studied; however, the cost of nano synthesis by some of the above methods is often very high, and the use of toxic materials in different stages is rugged synthetic that affects human health and is not friendly to the environment [1]. Therefore, there is an urgent need to research and develop green biological methods to synthesize NPs. In addition, in nanotechnology, many studies have focused on synthesizing two metals, gold and silver, but there are few studies on synthesizing CuNPs, especially green synthesis.

With cheap synthesis cost, ease of use, large-scale application, and outstanding properties, especially antibacterial properties similar to precious metals, CuNPs have been and are being researched by manufacturers and researchers. Not only that, in solutions, bare nanoparticles are thermoelectrically unstable and tend to aggregate, losing their specific properties. Therefore, investigating different types of coatings for nano synthesis is also of interest to researchers. Typically, the research of [2] has successfully synthesized copper (I) oxide nanoparticles with the smallest particle size of 222 ± 13 nm, with the use of reducing agent, glucose, and polyvinyl alcohol (PVA) encapsulation showed the results that copper (I) oxide nanoparticles had the highest ability to kill E.

coli at a concentration of 160 ppm with a treatment time of 1 hour and a pH value of 8. Recently, [3] demonstrated antibacterial and antifungal activity of copper oxide/carbon (CuO/C) nanocomposites against Pseudomonas aeruginosa, Escherichia coli, Klebsiella neumonia, Staphylococcus aureus, Candida albicans, and Aspergillus niger. On the other hand, following the progress of science and technology, finding more optimal preservation methods for agricultural products such as fruits is receiving much attention, including the research direction of biopolymer films, the most prominent being edible films. With those mentioned superior antibacterial properties of CuNPs, along with the constant increase of pathogenic bacteria that threaten the lives of humans and other organisms, the research and application of CuNPs in life is significant and is a new and urgent direction.

For the above reasons, we decided to choose the research topic with the research content “Effects of capping agent concentration and reaction time on antimicrobial activities of copper nanoparticles (CuNPs)” 1. Thesis objects In this study, copper(I) oxide nanoparticles were synthesized by chemical reduction, using copper sulfate anhydrous as a precursor, D-Glucose as a reducing agent, and starch as a capping agent. We focused on investigating the factors affecting the antibacterial ability of copper nanoparticle solutions, such as reaction time, the concentration of capping agent (starch), and storage conditions. From there, find the conditions for synthesizing nano copper with the most optimal antibacterial ability.

Limits and scope of the study CuNPs were prepared by chemical reduction method with copper sulfate anhydrous as precursor and D - glucose as reducing agent in the presence of soluble starch capping agent. We investigated the influence of reaction time, encapsulation concentration, and nano solution storage conditions to form the optimal synthesis formula and process to obtain the nano solution with the best antibacterial activity. The bacteria used were Escherichia coli and the mold Colletotrichum gloeosporioides. Research content In this study, we conducted the following: - Synthesis of copper(I) oxide nanofluids - Investigate the effect of starch capping agent concentration on the antibacterial ability of copper(I) oxide nanofluids.

- The effect of copper concentration on antimicrobial resistance was investigated. - Evaluation of the size, distribution, and morphology of copper nanoparticles by TEM, SEM, and Zeta potential. The scientific and practical significance of the topic Nanotechnology has promoted development in all fields, especially biomedical, energy, environment, information technology, military,etc. Nano copper(I) oxide with good antibacterial ability is a potential material for food preservation.

Applying active film to preserve food products such as meat, fish, fruit, etc., prolongs the storage time and minimizes pathogenic microorganisms' growth.
